    Lyngbya majuscula Blooms in an Enclosed Marine Environment

    Cyanobacterial blooms are a cause of concern because of their potential impacts on the marine environment. In Sentosa Cove, Singapore, Lyngbya majuscula blooms appeared regularly in the highly enclosed boat canals traversing the seafront residential development. This study investigated whether sediments resuspended by physical disturbance liberated nutrients that contribute to the blooms. Sediment resuspension events were mimicked in containers of sediment collected from the canals. Lyngbya majuscula that were incubated in containers with resuspended sediment attained greater biomass than those in filtered seawater only. Levels of iron, phosphates and nitrites in seawater with resuspended sediments were significantly higher than in those without. The results indicate that recurrent L. majuscula blooms in Sentosa Cove could be attributed to nutrient loading from sediment resuspension

    Seasonal variation of Sargassum ilicifolium (Phaeophyceae) growth on equatorial coral reefs

    Temporal and spatial variations in Sargassum ilicifolium thallus density and length were investigated on equatorial coral reefs in Singapore from November 2011 to October 2012. Thalli density varied little throughout the year, however, we found strong seasonal patterns in thallus length and identified temperature as the significant driver. Sargassum ilicifolium reached maximum length in December (110.39 ± 2.37 cm) during periods of cooler water temperatures, and minimum length in May (9.88 ± 0.48 cm) during periods of warmer water temperatures. Significant spatial variation was also observed for both thallus density and length of S. ilicifolium among reefs. Within reefs, densities of S. ilicifolium were higher on reef flats (20.40 ± 0.40 individuals · 0.25 m−2) compared to upper reef slopes (5.66 ± 0.23 individuals · 0.25 m−2). Our findings highlight that marked seasonality in the growth of canopy-forming macroalgae can occur within equatorial reef systems where temperature ranges are restricted (\u3c3°C)

    Could the wild population of Large Yellow Croaker Larimichthys crocea (Richardson) in China be restored? A case study in Guanjingyang, Fujian, China

    Over 90% of the world's fisheries have been fully exploited or over-fished. Included is the large yellow croaker (Larimichthys crocea), an important commercial fish species in China whose population was nearly depleted prior to the 1980s. Although overfishing and natural resources collapse present a daunting issue, some studies indicate that improved management strategies could aid in natural stock restoration to prevent depletion. We developed an integrated assessment method grounded on an ecosystem-based approach and deigned an integrated index with three key aspects of habitat suitability, natural population status and government & social interventions, to evaluate the potential restoration capacity of the species in a designated “national aquatic germplasm resource protected area” in Guanjinyang based on a data set spanning 1987 to 2015. The results show that although restoration efforts on research and rehabilitation have increased greatly since late 1990s, the effectiveness stays moderate and the natural population remains near depletion

    Reefs at Risk: A Map-Based Indicator of Threats to the Worlds Coral Reefs

    This report presents the first-ever detailed, map-based assessment of potential threats to coral reef ecosystems around the world. "Reefs at Risk" draws on 14 data sets (including maps of land cover, ports, settle-ments, and shipping lanes), information on 800 sites known to be degraded by people, and scientific expertise to model areas where reef degradation is predicted to occur, given existing human pressures on these areas. Results are an indicator of potential threat (risk), not a measure of actual condition. In some places, particularly where good management is practiced, reefs may be at risk but remain relatively healthy. In others, this indicator underestimates the degree to which reefs are threatened and degraded.Our results indicate that:Fifty-eight percent of the world's reefs are poten-tially threatened by human activity -- ranging from coastal development and destructive fishing practices to overexploitation of resources, marine pollution, and runoff from inland deforestation and farming.Coral reefs of Asia (Southeastern); the most species-rich on earth, are the most threatened of any region. More than 80 percent are at risk (undermedium and high potential threat), and over half are at high risk, primarily from coastal development and fishing-related pressures.Overexploitation and coastal development pose the greatest potential threat of the four risk categories considered in this study. Each, individually, affects a third of all reefs.The Pacific, which houses more reef area than any other region, is also the least threatened. About 60 percent of reefs here are at low risk.Outside of the Pacific, 70 percent of all reefs are at risk.At least 11 percent of the world's coral reefs contain high levels of reef fish biodiversity and are under high threat from human activities. These "hot spot" areas include almost all Philippine reefs, and coral communities off the coasts of Asia, the Comoros, and the Lesser Antilles in the Caribbean.Almost half a billion people -- 8 percent of the total global population -- live within 100 kilometers of a coral reef.Globally, more than 400 marine parks, sanctuaries, and reserves (marine protected areas) contain coral reefs. Most of these sites are very small -- more than 150 are under one square kilometer in size. At least 40 countries lack any marine protected areas for conserving their coral reef systems

    Short Term Exposure to Heat and Sediment Triggers Changes in Coral Gene Expression and Photo-Physiological Performance

    Corals, together with their endosymbiotic Symbiodiniaceae, are known to exhibit a suite of mechanisms for survival under stressful conditions. However, exactly how the host animal responds to varying environmental conditions remains unclear. In this study, we tested two relevant environmental factors that can have deleterious effects on corals: heat and sediment. We examined among-genotype responses of Pocillopora acuta to these factors with RNA-Seq in concert with widely-used tools for assessing the physiological conditions of corals. Heat and sediment treatments were applied in a 2 × 2 crossed experimental design: (1) ∼30°C without sediment (control, “C”), (2) ∼30°C with sediment (sediment-only, “S”), (3) ∼32°C without sediment (heat-only, “H”), (4) ∼32°C with sediment (heat + sediment, “H+S”) over four consecutive days (3-h daily exposure) in ex situ aquarium conditions. A clear differentiation in gene expression patterns was observed in corals exposed to heat alone and to heat with sediment, relative to the control treatment. Few transcripts (∼3) were found to be differentially expressed for corals exposed to sediment only. The greater impact of heat was supported by photo-physiological measurements that showed significant effects on maximum quantum yield and average symbiont density among genotypes of P. acuta. The combined effect of heat and sediment caused a greater reduction in average symbiont density than the effect of sediment alone. Furthermore, “H+S” disrupted the ability of corals to maintain processes involving assembly and disassembly of cilium which suggests a synergistic effect between the two factors. We also found that host-specific genes which were expressed differentially may not be related to their interactions with algal symbionts. Rather, these genes are involved in a variety of biological functions including, but not limited to, cilium biogenesis and degradation, cilia motility, innate and adaptive immune responses, cell adhesion and bone mineralization, and processes involved in the cell cycle. These results reflect the complex response of the host alone. Overall, our findings indicate that acute heat stress in tandem with sediment can depress photo-physiological performance and trigger considerable changes in host gene expression

    Contrasting Patterns of Coral Bleaching Susceptibility in 2010 Suggest an Adaptive Response to Thermal Stress

    Background: \ud Coral bleaching events vary in severity, however, to date, the hierarchy of susceptibility to bleaching among coral taxa has been consistent over a broad geographic range and among bleaching episodes. Here we examine the extent of spatial and temporal variation in thermal tolerance among scleractinian coral taxa and between locations during the 2010 thermally induced, large-scale bleaching event in South East Asia.\ud \ud Methodology/Principal Findings: \ud Surveys to estimate the bleaching and mortality indices of coral genera were carried out at three locations with contrasting thermal and bleaching histories. Despite the magnitude of thermal stress being similar among locations in 2010, there was a remarkable contrast in the patterns of bleaching susceptibility. Comparisons of bleaching susceptibility within coral taxa and among locations revealed no significant differences between locations with similar thermal histories, but significant differences between locations with contrasting thermal histories (Friedman = 34.97; p,0.001). Bleaching was much less severe at locations that bleached during 1998, that had greater historical temperature variability and lower rates of warming. Remarkably, Acropora and Pocillopora, taxa that are typically highly susceptible, although among the most susceptible in Pulau Weh (Sumatra, Indonesia) where respectively, 94% and 87% of colonies died, were among the least susceptible in Singapore, where only 5% and 12% of colonies died.\ud \ud Conclusions/Significance: \ud The pattern of susceptibility among coral genera documented here is unprecedented. A parsimonious explanation for these results is that coral populations that bleached during the last major warming event in 1998 have adapted and/or acclimatised to thermal stress. These data also lend support to the hypothesis that corals in regions subject to more variable temperature regimes are more resistant to thermal stress than those in less variable environments

    Urban coral reefs: Degradation and resilience of hard coral assemblages in coastal cities of East and Southeast Asia

    © 2018 The Author(s) Given predicted increases in urbanization in tropical and subtropical regions, understanding the processes shaping urban coral reefs may be essential for anticipating future conservation challenges. We used a case study approach to identify unifying patterns of urban coral reefs and clarify the effects of urbanization on hard coral assemblages. Data were compiled from 11 cities throughout East and Southeast Asia, with particular focus on Singapore, Jakarta, Hong Kong, and Naha (Okinawa). Our review highlights several key characteristics of urban coral reefs, including “reef compression” (a decline in bathymetric range with increasing turbidity and decreasing water clarity over time and relative to shore), dominance by domed coral growth forms and low reef complexity, variable city-specific inshore-offshore gradients, early declines in coral cover with recent fluctuating periods of acute impacts and rapid recovery, and colonization of urban infrastructure by hard corals. We present hypotheses for urban reef community dynamics and discuss potential of ecological engineering for corals in urban areas

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data
